There is no way to manually change the landing altitude as it is correlated with the origin and destination airports in the FMC. The origin altitude will be used in the event of an air turn back early in the flight.
Are you trying to use your flight simulator to do touch and goes at some airport which is neither the origin or destination in the flight plan? That may result in the EICAS advisory message you’ve described.

Yes you can. The 777, and all other Boeing models, has a Landing Altitude (LDG ALT) Selector on the overhead panel that lets you manually select a landing altitude for pressurization.
